General description

Myelin-oligodendrocyte glycoprotein (UniProt: Q16653) is encoded by the MOG gene (Gene ID: 4340) in human. Myelin oligodendrocyte glycoprotein (MOG) is a type I integral membrane protein predominantly expressed in the central nervous system (CNS), specifically on the outermost surface of myelin sheaths and oligodendrocyte membranes. MOG is a member of the immunoglobulin superfamily, containing an extracellular Ig variable domain that adopts a beta-sandwich conformation. MOG plays a critical role in myelin sheath formation, maintenance, and integrity, as well as in cell-cell communication within the CNS. It is thought to act as an adhesion molecule, contributing to the structural stability of myelin. MOG also serves as a target for immune responses, particularly in inflammatory demyelinating diseases such as multiple sclerosis (MS) and MOG antibody-associated disease (MOGAD). Pathogenic mechanisms involve complement-dependent cytotoxicity and antibody-mediated damage, leading to demyelination and inflammation. MOG′s expression is developmentally regulated, appearing late during oligodendrocyte maturation. It is highly localized to the surface of myelin and oligodendrocyte processes, enabling its role in immune-mediated diseases. Application
Quality Control Testing
Evaluated by Western Blotting in mouse brain tissue lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected MOG in mouse brain tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected MOG in human and rat brain tissue lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected MOG in human cerebral cortex tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound with MOG peptide with at least one billion-fold (1,000,000,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2I9 is a ZooMAb® rabbit recombinant monoclonal antibody that detects MOG. It targets an epitope within 25 amino acids from the N-terminal extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
