ACE (ACE-Okra Glow), a non-fluorescent molecular rotor, serves as the fluorogenic ligand for the Okra RNA aptamer (more than 400-fold fluorescence enhancement), forming the Okra505 complex that exhibits exceptional brightness and photostability in live-cell imaging. The Okra-ACE system demonstrates rapid binding kinetics (KD ≈ 1.2 nM; kon = 0.97 × 106 M−1 s−1 & koff = 1.0 × 10-3 s−1), low overall background fluorescence, pH resistant and good cell permeability, enabling robust visualization of mRNA dynamics, including detection of low-abundance mRNAs and observation of mRNA recruitment to subcellular structures like stress granules. Compatible with standard GFP filter sets, Okra505 allows for dual-color super-resolution imaging when combined with other fluorescent RNA aptamers, facilitating detailed studies of mRNA behavior and interactions in living cells while accounting for potential lysosomal localization. Excitation ~ 468 nm, emission ~ 505 nm & Φ = 0.93.
