Transthyretin (TTR) tetramer stabilizer.
Tafamidis is a transthyretin (TTR) tetramer stabilizer that binds selectively and with negative cooperativity to the two normally unoccupied thyroxine-binding sites of the tetramer (Kd ~2 nM and ~200 nM), kinetically stabilizing the wild-type as well as the variant TTR complexes (V30M, F64S, Y69H, I84S, V122I, and L111M) and preventing their dissociation into fibril-forming monomers.
