Nek1 Inhibitor 10f may be used to analyze the effects of NEK1 inhibition on ciliogenesis in zebrafish pronephros. It may also be used to evaluate its efficacy, toxicity, and bioavailability in models of zebrafish polycystic kidney disease.
Biochem/physiol Actions
Nek1 Inhibitor 10f (Nek1i-10f) is a cell-permeable azaindolo-D-tryptophanyl compound that acts as a potent and ATP-competitive Nek1 selective inhibitor (IC50 at 10 µM ATP = 330 nM; 96% inhibition at 1 µM). Nek1 Inhibitor 10f is shown to dominantly affect IKKb activity in a 50-kinase panel. Nek1i-10f is bioavailable, highly efficacious with less toxicity and causes cystic kidneys in zebrafish embryos (10 µM). Nek1-IN-10f reduces NEK1 levels and perturbs nucleocytoplasmic transport in induced pluripotent stem cell motor neurons.
