StraightFrom® Whole Blood and Bone Marrow CD138 MicroBeads have been developed for the positive selection of CD138+ cells directly from anticoagulated whole blood or bone marrow by using the autoMACS® Pro Separator or the Whole Blood Column Kit. No sample preparation is required, including density gradient centrifugation or erythrocyte lysis.
The CD138 antigen, also known as syndecan-1, is primarily found on normal and malignant plasma cells in the bone marrow. In the peripheral blood of healthy donors approximately 50% of plasma cells express this antigen. The CD138 antigen is not expressed by naive B cells, germinal center B cells, memory B cells, T cells, or monocytes.
Anticoagulated cell samples of 0.25–15 mL are labeled with StraightFrom® Whole Blood and Bone Marrow CD138 MicroBeads and subsequently separated with the autoMACS® Pro Separator or using the Whole Blood Column Kit. StraightFrom Whole Blood and Bone Marrow MicroBeads in combination with the autoMACS Pro Separator allow standardization of the cell separation procedure and safe handling of hazardous samples.
Isolation of CD138+ cells from whole blood or bone marrow. The purified CD138+ cells are well suited for further flow cytometric, functional, or molecular analysis such as FISH, PCR analysis, microarrays, and protein analysis, and for analysis of cell surface markers or intracellular antigens, for example immunoglobulin light chains.
