AMXT1501 is a lipophilic polyamine analog that acts as a potent polyamine transport inhibitor. When combined with the polyamine biosynthesis inhibitor DFMO, AMXT1501 potently inhibits cell growth in cancer cultures (GI50 in nM in the presence of 5 mM DFMO, 500 nM spermidine & 1 mM aminoguanidine = 76/MDA-231, 120/PC-3, 31/A375, 170/SK-OV-3) and induces complete or nearly complete remission of cutaneous squamous cell carcinomas (SCC) growth in a transgenic ornithine decarboxylase (ODC) mouse model of skin cancer in vivo (0.5-0.7 mg/kg via b.i.d. i.p. with DFMO at 0.5% in drinking water).
