Iodoacetamide-alkyne C6 (IAA-C6, IA-alkyne-C6, IAM-alkyne-C6) is a bifunctional thiol-reactive probe designed for activity-based protein profiling (ABPP) of reactive cysteine residues in live cells. IA-alkyne probe combines an iodoacetamide group (covalent warhead) for cysteine labeling with an alkyne moiety that serves as a click-chemistry handle, enabling subsequent attachment of fluorophores or biotin-linked reporter for visualization or enrichment, respectively. Optimal usage concentrations are ≤3 μM for cellular studies and ~100 μM for lysates and tissue homogenates, facilitating comprehensive proteomic analysis of cysteine-dependent enzyme activities and protein functions.
