Perampanel is an orally active, noncompetitive AMPA receptor antagonist that inhibits 2 μM AMPA-, but not NMDA-, induced Ca2+ in cultured rat cortical neurones (IC50 = 93 nM) and display therapeutic efficacy in murine models of epilepsy in vivo (audiogenic, MES-induced, and PTZ-induced seizures in mice ED50 = 0.47, 1.6, and 0.94 mg/kg p.o., respectively) and a rat amygdala-kindling model (1-10 mg/kg p.o.).
