General description
Heat shock factor protein 1(UniProt: Q00613; also known as HSF 1, Heat shock transcription factor 1, HSTF 1) is encoded by the HSF1 (also known as HSTF1) gene (Gene ID: 3297) in human. HSF1 is a stress-inducible transcription factor that plays a key role in the transcriptional activation of the heat shock response (HSR), leading to the expression of a large class of molecular chaperones heat shock proteins (HSPs). In unstressed cells, it is present in cytoplasm in its monomeric, inactive form and homotrimerizes in response to stress and translocates to the nucleus where it binds to heat shock element (HSE) sequences in promoter of heat shock protein genes and acquires transcriptional ability. In unstressed cells, its spontaneous homotrimerization is inhibited by intramolecular interactions between the hydrophobic repeat HR-A/B and HR-C regions. HSF1 also has several other functions independently of its transcriptional activity. It is reported to be involved in the repression of Ras-induced transcriptional activation of the c-Fos gene in heat-stressed cells. It is also involved in stress-induced cancer cell proliferation in a IER5-dependent manner. In unstressed cells, it remains phosphorylated on multiple serine and threonine residues. These phosphorylations are constitutive and are implicated in the repression of HSF1 activity. Its phosphorylation on serine 121 by MAPKAP kinase inhibits HSF1 homotrimerization, DNA-binding and transactivation activities. Phosphorylation on serine 303 by GSK3-β and on serine 307 by MAPK3 within the regulatory domain represses HSF1 transcriptional activity and increases its nuclear export in a CRM1 manner. Phosphorylation on serine 230 by CAM kinase 2 and on threonine 142 by casein kinase 2 is reported to enhance its transactivation activity upon heat shock. Application
Quality Control Testing
Evaluated by Western Blotting in MCF-7 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HSF1 in MCF-7 cell lysate.
Tested applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HSF1 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound HSF1 peptide with a KD of 7.7 x 10-6 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HSF1 in human kidney tissue sections.

Clone 1J20 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Heat shock factor 1 (HSF1). It targets an epitope within 21 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
