HLA class I histocompatibility antigen, B alpha chain (UniProt: P01889; also known as Human leukocyte antigen B, HLA-B) is encoded by the HLA-B (also known as HLAB) gene (Gene ID: 3106) in human. HLA-B is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 25-309), a transmembrane domain (aa 309-333), and a cytoplasmic domain (aa 334-363). It is an antigen-presenting major histocompatibility complex class I (MHCI) molecule that in complex with β2-microglobulin displays viral and tumor-derived peptides on antigen-presenting cells for recognition by -β T cell receptor (TCR) on HLA-B-restricted CD8+ T cells. This helps antigen-specific T cell immune response to eliminate infected or transformed cells. It typically presents intracellular peptide antigens of 8 to 13 amino acids that arise from cytosolic proteolysis via constitutive proteasome and interferon- -induced immunoproteasome. It contains three alpha ( 1, 2, and 3) domains. Residues 101-107 determine Bw4/Bw6 motifs, which serologically distinguish HLA-B alleles 1 domain (aa 25-114) is a structural part of the peptide-binding cleft and residues 101-107 in this region determine Bw6 motif that serologically distinguish HLA-B alleles. 2 domain (aa 115-206) is reported to mediate the interaction with Transporter Associated with Antigen Processing ( TAP1-TAP2) complex. 3 domain (aa 207-298)mediates the interaction with CD8 co-receptor. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human HLA-B.


Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HLA-B in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected HLA-B in lysates from Ramos cells and Human tonsil tissue and 1:1,000 dilution in A431 cells.

Affinity Binding Assay: A representative lot of this antibody bound HLA-B peptide with a KD of 7.1 x 10-8 in an affinity binding assay.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected HLA-B in Human tonsil tissue sections.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected HLA-B in one million Human peripheral blood mononuclear cells (PBMC).

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HLA-B in Human tonsil tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.


Biochem/physiol Actions

Clone 1L21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects HLA-B. It targets an epitope within 25 amino acids within the extracellular domain from the C-terminal half.


Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.


Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
