General description
Signal-induced proliferation-associated protein 1 (UniProt: Q96FS4; also known as Sipa-1, GTPase-activating protein Spa-1, p130 SPA-1) is encoded by the SIPA1 (also known as SPA1) gene (Gene ID: 6494) in human. SIPA1 is a GTPase activator for the nuclear Ras-related regulatory proteins Rap1 and Rap2 that negatively regulates Rap via hydrolysis of active GTP-Rap to inactive GDP-Rap. It is mostly localized in the perinuclear membranous region, co-locating with Rap1 and Rap2. It is expressed in fetal and adult tissues and is abundantly expressed in thymus, spleen, peripheral blood lymphocytes, and spinal cord. Three major domains are identified in SIPA1: Rap-GAP domain (aa 321-539); PDZ domain (aa 687-763) and a coiled coil domain (aa 972-1034). SPA-1 is involved in the metastasis of various solid tumors and is indicative of a poor prognosis. In MDA-MB-231 cells SIPA1 is mainly localized in the nucleus and knockdown of SIPA1 gene reduces adhesion, migration, and invasion in these cells and reduces their metastatic capacity. However, it promotes these cells to proliferate. Nuclear SIPA1 is shown to interact with and activate integrin β1 promoter in breast cancer cells. Application
Quality Control Testing
Evaluated by Western Blotting in Mouse spleen tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SIPA1 in Mouse spleen tissue lysate.
Tested Applications.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SIPA1 in Human tonsil tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound SIPA1 peptide with a KD of 2.6 x 10-7 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SIPA1 in Jurkat and THP-1 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1D5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Signal-induced proliferation-associated protein 1 (SIPA1).
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
