pro-ADM (UniProt: P35318; also known as Mid-regional pro-adrenomedullin) is encoded by the ADM (also known as AM) gene (Gene ID: 133) in human. ADM is synthesized with a signal peptide (aa 1-21) and is cleaved into Adrenomedullin (AM; aa 95-146) and Proadrenomedullin N-20 terminal peptide (PAMP; aa 21-41). AM and PAMP serve as potent hypotensive and vasodilatory agents involved in physiologic control of fluid and electrolyte homeostasis. In the kidney, AM acts as a diuretic and natriuretic, and both AM and PAMP are known to inhibit aldosterone secretion. ADM contains a region known as mid-region ProADM (aa 45-92) that lacks any activity but is a reliable biomarker for sepsis and progression to septic shock. ADM has been found in all epithelial surfaces that separate the internal and external environment, such as integument, respiratory, digestive and genito-urinary tract. It is also found in all bodily secretions and imparts protection against external pathogens. Its highest levels are reported in pheochromocytoma and adrenal medulla. ADM as well as PAMP also display potent antimicrobial action against both Gram-positive and Gram-negative bacteria. MR-proADM, a fragment of 48 amino acids that splits from the proADM and is present in a ratio of 1:1 with ADM, proportionally represents the levels and activity of ADM. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. KLH-conjugated linear peptide corresponding to 24 amino acids from the propeptide N-terminal half of human ADM.
Application
Quality Control Testing
Evaluated by Western Blotting with recombinant Human ADM Protein.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant human ADM protein.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MR-proADM in human placenta tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound MR-proADM peptide with a KD of 9.9 x 10-7 in an affinity binding assay.

Anti-MR-proADM, clone 2B19 ZooMAb®, Cat. No. ZRB1182, is a recombinant Rabbit monoclonal antibody that specifically targets MR-proADM and is tested for use in Affinity Binding Assay, Immunohistochemistry (Paraffin), and Western Blotting.
Biochem/physiol Actions
Clone 2B19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human ADM. It targets an epitope within the mid-region (MR) of ADM.
Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
