General description
TGF-beta receptor type-1 (UniProt: P36897; also known as EC:2.7.11.30, TGFR-1, Activin A receptor type II-like protein kinase of 53kD, Activin receptor-like kinase 5, ALK-5, Serine/threonine-protein kinase receptor R4, SKR4, Transforming growth factor-beta receptor type I, TbetaR-I) is encoded by the TGFBR1 (also known as ALK5, SKR4) gene (Gene ID: 7046) in human. TGF-β receptor type-1 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 34-126), a transmembrane domain (aa 127-147), and a cytoplasmic domain (aa 148-503). It is widely expressed and is most abundant in placenta and least abundant in brain and heart. It is also expressed in a variety of cancer cell lines. Application
Quality Control Testing
Evaluated by Western Blotting with IgG-His-tagged recombinant protein fragment from the extracellular domain of Human TGFβ receptor 1.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IgG-His-tagged recombinant fragment corresponding to 100 amino acids from the extracellular domain of Human TGFβ receptor 1.
Tested Applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TGF β receptor 1 in HepG2 cells.
Affinity Binding Assay: A representative lot of this antibody bound TGF β receptor 1 peptide with a KD of 1.2 x 10-7 in an affinity binding assay.
Enzyme Immunoassay (ELISA) Analysis: Serial dilutions from a representative lot detected hIgG-His-tagged recombinant fragment corresponding to 100 amino acids from the extracellular domain of Human TGF β receptor 1.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 4H18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects TGF-β receptor type 1 (TGF β-R1). It targets an epitope within 16 amino acids from the extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
