SerBut (O-butyryl-L-serine) is an orally available, serine-conjugated butyrate precursor with therapeutic efficacy against autoimmune arthritis and neuroinflammation in vivo in mouse models of collagen antibody-induced arthritis (CAIA) (anti-collagen mAb on day 0, LPS i.p. on day 3, daily 20 mg SerBut/mouse p.o. starting on day -14 or twice daily 25 mg SerBut/mouse p.o. starting on day 4) and experimental autoimmune encephalomyelitis (EAE) (MOG35-55/CFA + PTX on day 0, daily 24 mg SerBut/mouse p.o. starting on day 2 with or twice daily 24 mg SerBut/mouse p.o. starting on day 2 without 100 mM SerBut in drinking water throughout starting day -14).
